Multilevel analyses tested the association between CRF or MF and cognitive outcomes while accounting for the influence of the other fitness variable and relevant demographic factors. Results: CRF predicted response accuracy during incongruent flanker trials, the condition requiring greater inhibition. For the working memory task, CRF predicted greater target accuracy and greater d' scores on the 1-back task, requiring lesser amounts of working memory. In the 2-back task, which requires greater amounts of working memory, CRF also predicted greater target and non-target accuracy and d' scores. Comparatively, MF did not predict any cognitive outcomes after adjustment for CRF. Conclusion: CRF was selectively related to better performance during task conditions that require greater amounts of inhibition and working memory. This finding suggests that CRF, but not MF, may benefit cognitive control in older adolescents. Two of the 23 identified CpGs (cg00253568 (FTO) and cg00213123 (CYP1A1)) were associated with either TG (males and females), diastolic blood pressure (females only) or HDL-C (males only), after Bonferroni correction. Discussion: This study demonstrates a critical timing of cigarette smoke exposure over the life-course for establishing persistent changes in DNA methylation into adolescence in a dose dependent manner. There were significant associations between offspring CpG methylation and adolescent cardiovascular risk factors, namely TG, HDL-C and diastolic blood pressure. From the age of 13, males were significantly less likely to have a diagnosis of MDD than females with the maximum sex difference occurring at the age of 15 (OR 0.24, 95% CI 0.13–0.47). All lifestyle risk factors were at some point significantly associated with MDD, but these associations did not differ by sex, except for body mass index. Discussion: These findings suggest that interventions designed to prevent the development of depression should be implemented in early adolescence, ideally before or at the age of 13 and particularly among young females given that the prevalence of MDD begins to rise and diverge from young males. Clinically meaningful cut-offs for disordered eating behaviours were generated for analysis. Results: Disordered eating behaviours, specifically self-induced vomiting (diabetes 19.2%, no diabetes 3.3%; p <  0.001), laxative use (diabetes 15.4%, no diabetes 2.1%; p <  0.001), use of cigarettes (diabetes 26.9%, no diabetes 4.3%; p <  0.001) and other drugs (diabetes 28.9%, no diabetes 4.0%; p <  0.001), cleanse/detox (diabetes 30.8%, no diabetes 10.5%; p <  0.001) and extreme weight loss diets (diabetes 13.5%, no diabetes 4.7%; p <  0.003) were higher in those reporting a diagnosis of diabetes. In addition, 17% of those with diabetes reported frequent insulin restriction (≥ once per week), and insulin restriction was associated with more frequent disordered eating behaviours. Conclusion: There was a high rate of disordered eating behaviours in adolescents with diabetes compared to their peers without diabetes. The findings of this study may have the potential to inform future health promotion, prevention, and early intervention approaches for those with comorbid diabetes and disordered eating behaviours.
